Several Total Wars I would like to see:
Civil War: Total War WW1: Total War
The Persian Wars The Long War.
I would also like to see something take place in Africa. Maybe the Zulu Wars or the Boer Wars.

Earth: Total War
Evolve through the ages, develop technologies nobody has thought of before, make your own progress through time and lead your people to glory. Follow history in the small details, or make your own, to your liking. Meet new civilizations, trade, do diplomacy, or wage TOTAL WAR!
Freedom is overrated.

As most everyone is chiming in, I might as well too.
I like the idea of an updated RTW. But how about thinking outside the box (sort of)? The Third Age: Total War was an excellent mod. If the CA were to made an actual game like TATW, imagine what that would look like?

I was disappointed when it came to empire TW and Napoleon, for me the muskets and cannon artillery ruined it.. I have longed for a Shogun 2.

The people calling for the WWi/II setting are now becoming a small minority. Before Empire everybody wanted the guns and gunpowder in droves but now that Empire and Napoleon released, the people see it's not that easy for CA to make a game with almost purely ranged combat. The AI seemed dumber than ever before and having to wait for every one of your squad to be in position before they start firing could be disastrous at times.
I'm all for Rome 2 and undoubtedly that is the next step. I'll even take a Medieval 3 if it means upgraded graphics and settings.
