No, just like other Live titles, you can use an offline profile.

Being logged in to an online Live account is the only way to earn the Achievements and get access to the Scarecrow Challenge map if you got the game from GameStop, since you have to redeem a code through Live in order to download it. But like PWizard said, you don't need an online account to play.
