First please make sure you got the latest version. There has been some bugs in previous versions where the editor wouldn't work in certain situations. Then I've made a video showing how to use the editor to replace items, changing quantity or spawn new items on empty slots. Hope it helps:
So I have the latest version, I'm in the same room as the thumbnail from the video - I alt/tab out of game, inventory noticeably disappears in game, I make a change, and nothing happens. I can't watch your video because it never loads (I am ethernet cable plugged in and no problems being online, I can load most things instantly with my 500 mbps speeds, so not sure what the issue is there...?). Nothing changes when I select something alternate. Any help would be appreciated!

I tried the Infinite Combat knife, and I put it in the chest. I saved the game went out to find the Diamond key, had to reload and then gone.
Do we not get to keep the item(s)? Or do we have to always have to keep re-adding it to the inventory?

Just a question that might help them a bit, have you experienced this with ALL items, or just certain unlockable ones like the unbreakable knife/etc? The first thing I'd suspect is that the game is detecting an item that it doesn't show as unlocked/enabled yet, and disabling/removing it as a result, but if it's happening with, say, ammunition, then there are other possibilities.
For what it's worth, I haven't had that happen, but I've tended to use it more on things I kept in my inventory (ammo, wooden boards, etc), I'm not sure I've deposited anything except excess ammo.
