Must I play the game in multiplayer mode? When I play as a single player, I cannot load any saved checkpoint.

There is no going back to check points once you've ended a mission, in SP and MP.

Why there is no load/continue game options for this game. Also, I alway starts from the beginning when Iopen up S3.

No, you don't. Once you finish an area, it turns from black and white to colour. You have to follow the given path or you can just skip everything since it is not required to unlock something to progress. The recommended levels makes sense though.
You just choose your savegame at the beginning.
