I can't seem to figure out what this 43rd weapon is...it's the last one I need to get, but it's not in the shops, and I've done everything else.

If you have the Deluxe Edition, activate it and you'll get the Tribal Knife and a cool sniper rifle right from the beginning.
Also, there are the signature weapons, but I think you already have them.

according to that list I have everything, but I'm still at only 42/43 weapons. I've checked the shop again as well and I own everything in it, so it has to be something that I can't buy, but what could it be?

Do you have the custom magnum? The one you get by redeeming Uplay award points or whatever they call it.

According to him, he has all the Signature weapons... The only thing I can remember now is the Japanese Tanto.

I have the tanto and the uplay gun too...this is really odd...I've done everything else, I've got all the relics, letters, and memory cards, I've finished the story, and have done all the missions, my progress is at 99%, just this missing weapon is all thats left.

HAAAAA.....you guys are forgetting one of the most secret weapons in the game, found only after watching the movie "Ted" and then playing the game.......the flash gordon water pistol...TA DAAAAAAAAA

Maybe the mechanic's tool? I don't know if that is a weapon or not, but if it is an you don't have it then...

Speaking about the mechanic's tool... You know, I've been bothered by not finding a way to change weapons from those in the little round weapon selector except by going to a machine or store and activating them there. Does anyone know a way to get all your weapons "online" so to speak so that you can get whatever one you need on the fly while you are out in the field? That would be handy. A couple of times I encountered guys with broken down cars and though I did have the tool in the "rucksack", I could not immediately produce it because I had other things in the 4 slots available in the game and didn't know how to get something different on the spot. So if anyone knows how to do that please reply.

PS... I'm also one of those ol' retired guys who spends his nights playing games, and although I used to fix computers for a living, some of this software eludes me. I'd call the inventory system in this game "Bysantine" at best.
